Herpes zoster (or simply zoster), commonly known as shingles and also known as zona, is a viral disease characterized by a painful skin rash with blisters in a limited area on one side of the body, often in a stripe. The initial infection with varicella zoster virus (VZV) causes the acute (shortlived) illness chickenpox, and generally occurs in children and young people. Once an episode of chickenpox has resolved, the virus is not eliminated from the body but can go on to cause shinglesan illness with very different symptomsoften many years after the initial infection. Herpes simplex virus type 1 (HSV1) establishes lifelong latent infection in sensory neurones, making it an ideal gene therapy vector to target neuronal cells. To study retrograde transport of HSV1, capsid and tegument proteins were tested for interactions with the motor cytoplasmic dynein, using a yeast twohybrid system and in vitro pull down assays. A strong interaction was demonstrated between the HSV1 outer capsid protein VP26 (UL35) and the homologous 14 kDa dynein light chains RP3 and Tctex1. The functional importance of VP26 for retrograde transport was confirmed by microinjecting recombinant HSV1 capsids into living cells. Capsids containing VP26 moved closer to the cell nucleus, while capsids without VP26 remained in a random distribution. Our results suggest that the HSV1 outer capsid protein VP26 mediates binding of incoming capsids to the retrograde motor cytoplasmic dynein during cellular infection, through interactions with dynein light chains. It is hoped that these findings will help in the development of a synthetic viral vector, which may allow targeted gene therapy in patients with neurological diseases. The microRNA (miRNA) and RNA interference (RNAi) pathways have recently emerged as an important aspect of virus-host cell interaction. This interaction can occur in several different ways and may favor either the virus or the host cell. Plants and invertebrates use RNAi as a first line of defense against virus infection by cleaving long, double-stranded viral transcripts into small interfering RNAs. However, it remains to be determined whether mammalian cells also initiate a similar response to infection. Here we present evidence that mammalian cells in fact do not induce an antiviral RNAi defense in response to infection by primate retroviruses. Viruses may also interact with host cells by encoding miRNAs to regulate either cellular or viral gene expression. Here we demonstrate that herpes simplex virus type 1 (HSV-1) encodes at least five miRNAs which are primarily expressed during latency. Two of these miRNAs modulate expression of viral genes required for productive replication. We hypothesize that down regulation of these viral genes by these latency associated miRNAs allows HSV-1 to establish and maintain the latent state.
